Studies of the High Molecular Weight Penicillin-binding Proteins of Bacillus subtiEs*
نویسنده
چکیده
Seven or eight penicillin-binding proteins (PBPs) were detected in Bacillus subtilis membranes. By introducing covalent affinity chromatography employing cephalosporins as ligands, milligram amounts of three high molecular weight PBPs (PBP 1 ab, M, = 120,000; PBP 2b, M, = 94,000; and PBP 4, M, = 78,000) were obtained without any contamination of the major PBP 5, the D-alanine carboxypeptidase. Small amounts of pure PBP 2b could be isolated by manipulation of the affinity chromatography conditions. Structural and physical properties of these proteins as well as the generation of one major penicilloyl peptide from each PBP by digestion with pepsin suggest that each PBP is the product of a separate gene. No enzymatic activity could be found in mixtures of these high molecular weight PBPs employing substrates used for the transpeptidase and D-alanine carboxypeptidase assays in particulate membrane fractions.

متن کاملSynthesis of peptidoglycan by high molecular weight penicillin-binding proteins of Bacillus subtilis and Bacillus stearothermophilus.
The high molecular weight penicillin-binding proteins (PBP(s) ) Bacillus subtilis PBPs 1, 2, and 4 and Bacillus stearothermophilus PBPs 1-4 were shown to catalyze peptidoglycan synthesis from the undecaprenol-containing lipid intermediate substrate in two assay systems. In a filter paper assay system, high levels of substrate polymerization occurred when reaction mixtures were incubated on What...
متن کاملTwo Proteins from Snake Venom have Potent Antibacterial Effects against Bacillus anthracis and Streptococcus pneumoniae
Background: Antibacterial proteins are widely expressed in snake venoms. Previously, we have isolated two immunodominant proteins with molecular weights of 14 and 65kD from the snake venom of Naja naja oxiana (N. oxiana). It was demonstrated that they had potent inhibitory effects against gram-positive bacteria, S. aureus and B. subtilis but were less effective against gram-negative bacteria, s...
متن کاملCharacterization of dacC, which encodes a new low-molecular-weight penicillin-binding protein in Bacillus subtilis.
The pbp gene (renamed dacC), identified by the Bacillus subtilis genome sequencing project, encodes a putative 491-residue protein with sequence homology to low-molecular-weight penicillin-binding proteins. Use of a transcriptional dacC-lacZ fusion revealed that dacC expression (i) is initiated at the end of stationary phase; (ii) depends strongly on transcription factor sigmaH; and (iii) appea...
متن کاملIdentification and characterization of pbpA encoding Bacillus subtilis penicillin-binding protein 2A.
Amino acid sequence analysis of tryptic peptides derived from purified penicillin-binding protein PBP2a of Bacillus subtilis identified the coding gene (now termed pbpA) as yqgF, which had been sequenced as part of the B. subtilis genome project; pbpA encodes a 716-residue protein with sequence similarity to class B high-molecular-weight PBPs. Use of a pbpA-lacZ fusion showed that pbpA was expr...
